ZF_TM_NubG

The ZF transmembrane Nub complex protein(ZM_TM_NubG) is a transmembrane protein with a complementary complex, the Zinc finger transmembrane cub complex (ZF_TM_Cub), that is designed to detect a specific DNA sequence. It is based on a membrane based yeast two hybrid system using a split ubiquitin. The membrane yeast two hybrid system is normally used to test for protein interaction between membrane proteins. When the ZF domains in the two complexes bind to their target DNA sequences on the same DNA molecule, the nub and cub domains are brought to close proximity activating the split ubiquitin, which releases a transcription factor.

ZF_TM_NubG protein contains multiple domains: signaling sequence domain, the Zinc finger domain, extracellular linker domain, transmembrane domain, B2 linker domain, and the NubG domain. The zinc finger domain in this complex binds to the sequence: TAGCAGCTG. The NubG domain is a part of the split ubiquitin used in membrane yeast two hybrid system. With the Zinc Finger nub and cub complexes, the binding to the target DNA sequence corresponds to a protein-protein interaction.
